The terminal elimination half-life of dutasteride is about 5 weeks at steady state (average 40 ng/ml). Following daily dosing of 0.5 mg, 65% of steady-state concentration is achieved after 1 month and approximately 90% after 3 months. Steady-state serum and semen concentrations are achieved at about 6 months. On average, semen concentrations are 11.5% of serum dutasteride concentrations in healthy patients at 1 year. Because of the long half-life of dutasteride, serum concentrations remain detectable (> 0.1 ng/ml) for up to 4 to 6 months after stopping therapy.
